If a game element is neither listed as a ranged, blast, burst, aura, etc., rather, it just states, "within x # of squares," does there need to be Line of Effect? For example, Rod of Corruption: "you can transfer your Warlock's Curse to each enemy within 5 squares of the original target." That is, does there need to be Line of Effect between the enemy and the original target?

You always need Line of Effect, unless the ability you're using states otherwise.
Another day, another three or four entries to my Ignore List.

You need Line of Effect from the Origin Space of the Effect, to be specific. In this case, the Origin Space would be the triggering creature and not yourself.
